Desyrel (trazodone hydrochloride)

Audience: Neuropsychiatric healthcare professionals.
FDA and Bristol-Myers Squibb notified healthcare
professionals of revisions to the CLINICAL PHARMACOLOGY
and PRECAUTIONS sections of the Desyrel labeling. Desyrel
is indicated for the treatment of depression. In vitro
drug metabolism studies suggest that there is a potential
for drug interactions when trazodone is given with the
CYP3A4 inhibitors ketoconazole, ritonavir, and indinavir.
It is likely that CYP3A4 inhibitors may lead to
substantial increases in trazodone plasma concentrations
with the potential for adverse effects. If trazodone is
used with a potent CYP3A4 inhibitor, a lower dose of
trazodone should be considered. Conversely, carbamazepine
reduced plasma concentrations of trazodone when
coadministered. Patients should be closely monitored to
see if there is a need for an increased dose of trazodone
when taken with carbamazepine.

Birth Control Shot Linked to Bone Thinning.

NEW YORK (Reuters Health) - Depo-Provera (depot
medroxyprogesterone), a popular birth control injection,
seems to promote bone loss, and the effects increase over
a 2-year period, new research shows.
Dr. Abbey B. Berenson, of the University of Texas Medical
Branch, Galveston, and colleagues compared the
bone-thinning effects of Depo-Provera, birth control
pills, and non-hormone contraception in a study of 191
young women.
Women who used Depo-Provera for 24 months had an average
loss in bone density of 5.7 percent. These patients
experienced a 3.2-percent loss between months 12 and 24.
Those who used pills, by contrast, had a 2.6-percent loss
at 24 months.  The results are published in the medical
journal Obstetrics and Gynecology.
Depo-Provera "is intended for long-term use," the authors
write. "Thus, additional studies that follow users of this
method for extended periods of time and those that
determine whether these changes are reversible are
critical so that physicians can accurately counsel
patients regarding this method."

Animal experiments have:
a 63% failure rate when detecting human carcinogens
a 75-95% failure rate for detecting drug side effects
a 70% failure rate for detecting drugs which cause birth defects
Success rates lower than those achieved by uneducated guesswork.
